In order for me to use Tiger's Roar, or something like it )flame artifact, frost artifact, etc) do I have to use a one handed weapon?

No but its bonus wont be added ontop of your stats if you have a two handed staff.

You can have an artifact with no other weapon or you can have an artifact equipped with a one handed weapon.
